چکیده

The stress activated protein kinases, Jun N-terminal kinase (JNK) and p38, orchestrate cellular responses to diverse environmental stresses and inflammatory signals. Crosstalk between JNK and p38 is emerging as an important regulatory mechanism in the inflammatory and stress responses. However, it is still unknown how this crosstalk regulates the signaling dynamics, cell-to-cell variabilities, and cell fate decisions at the single cell level. To address these issues, we established a multiplexed live-cell imaging system to simultaneously monitor JNK and p38 activities with high specificity and sensitivity at the single cell resolution. Exposure to various stresses and cytokines resulted in differential dynamics of JNK and p38 activation. In all cases, p38 negatively regulated JNK activity without affecting its own activation. We quantitatively demonstrate that p38 antagonizes JNK through both transcriptional and posttranslational mechanisms with the relative contribution of the pathway depending on the stimulant. This cross-inhibition of JNK appears to generate cellular heterogeneity in JNK activity in response to inflammatory cytokines and stresses.

Iranian crack induces hepatic injury through mitogen-activated protein kinase pathway in the liver of Wistar rat

Objective(s): Iranian crack (IC) is a heroin-based substance manifesting various pathologic side effects. Herein, we aimed to investigate the mechanism of IC-induced liver injuries in Wistar rats. Materials and Methods: Twenty male Wistar rats were randomly divided into two groups: control, and IC (0.9 mg/kg/day/IP, for 30 days). Mitochondrial reactive oxygen species (ROS) production was measur...

Inactivation of mitogen-activated protein kinase signaling pathway reduces caspase-14 expression in impaired keratinocytes

Objective(s):Several investigations have revealed that caspase-14 is responsible for the epidermal differentiation and cornification, as well as the regulation of moisturizing effect. However, the precise regulation mechanism is still not clear. This study was aimed to investigate the expression of caspase-14 in filaggrin-deficient normal human epidermal keratinocytes (NHEKs) and to explore the...

The Time Course of JNK and P38 Activation in Cerebellar Granule Neurons following Glucose Deprivation and BDNF Treatment

Low glucose condition induces neuronal cell-death via intracellular mechanisms including mitogen-activated protein kinases (MAPK) signaling pathways. It has been shown that low glucose medium decreases neuronal survival in cerebellar granule neurons (CGNs). In this study, we have examined the activation of JNK, p38kinase and ERK1/2 pathways in low glucose medium in CGNs. The CGNs were prepared ...

Dual specificity phosphatases 10 and 16 are positive regulators of EGF-stimulated ERK activity: Indirect regulation of ERK signals by JNK/p38 selective MAPK phosphatases

We have explored the possible role of dual specificity phosphatases (DUSPs) on acute EGF-mediated ERK signalling using high content imaging and a delayed MEK inhibition protocol to distinguish direct and indirect effects of the phosphatases on ERK activity. Using siRNAs, we were unable to find evidence that any of the MAPK phosphatases (MKPs) expressed in HeLa cells acts directly to dephosphory...
